Chloroquine is an aminoquinoline that is quinoline which is substituted at position 4 by a 5-diethylaminopentan-2-ylamino group at at position 7 by chlorine. It is used for the treatment of malaria, hepatic amoebiasis, lupus erythematosus, light-sensitive skin eruptions, and rheumatoid arthritis. CHLOROQUINE sulfate or phosphate oral Given that resistance of P. falciparum to chloroquine is widespread, this drug must not be used for the treatment of falciparum malaria in Africa, South America, Asia and Oceania. Unfortunately, within a decade of its introduction, P. falciparum parasite resistance to chloroquine was observed in most of the malaria-endemic countries. Nowadays, insurgence of resistance against chloroquine is a considerable hurdle for malaria control.

Ronald Ross discovered its transmission by mosquito in 1897. Alphonse Laveran was the first to identify the parasite in 1880, and named it Oscillaria malariae.

Since then, chloroquine resistance has spread to nearly all areas of the world where falciparum malaria is transmitted. Oct 01, 2018 Chloroquine resistance is widespread in P. falciparum and is reported in P. vivax. Before using Chloroquine for prophylaxis, it should be ascertained whether Chloroquine is appropriate for use in the region to be visited by the traveler. Chloroquine is administered orally as chloroquine phosphate. It also can be given by intramuscular injection as chloroquine hydrochloride. Chloroquine is effective against susceptible strains of the malarial parasites Plasmodium vivax, P. ovale, and P. falciparum, as well as certain parasitic worms and amoebas.
