Urinary excretion of chloroquine and its desethyl‐metabolite was monitored for up to 395 days after the last dose of oral chloroquine 300 mg base taken once weekly for 10 weeks as malaria prophyl. Very small amounts of chloroquine are excreted in breast milk; when given once weekly, the amount of drug is not sufficient to harm the infant nor is the quantity sufficient to protect the child from malaria. Chloroquine is rapidly and almost completely absorbed from the gastrointestinal tract. Excretion of chloroquine is quite slow, but is increased by acidification of the urine. Slightly more than half of the urinary drug products can be accounted for as unchanged chloroquine. Large i.m. injections or rapid i.v.

For all uses of chloroquine: WARNING/CAUTION: Even though it may be rare, some people may have very bad and sometimes deadly side effects when taking a drug. Do not start, stop, or change the dose of any drug without checking with your doctor.

Chloroquine was assayed in the semen of eight male volunteers aged between 19 and 45 years by a fluorimetric method before and after taking a total dose of 1.500 mg chloroquine base. The excretion of chloroquine and the major metabolite, desethylchloroquine, in breast milk was investigated in eleven lactating mothers following a single oral dose of chloroquine 600 mg base. The maximum daily dose of the drug that the infant can receive from breastfeeding was about 0.7% of the maternal start dose of the drug in malaria chemotherapy. The maximum daily dose of the drug that the infant can receive from breastfeeding was about 0.7% of the maternal start dose of the drug in malaria chemotherapy; therefore, separate chemoprophylaxis is required for the infant.
